In this important article, Dr. DeMarini reviews the history of the search for human germ-cell mutagens. He cites and describes the studies that have allowed him to conclude ionizing radiation, chemotherapy, smoking and air pollution are, in fact, human germ-cell mutagens. Through the use of advanced sequencing technologies, Dr. DeMarini anticipates molecular epidemiological analyses of parent-child cohorts will realize previously unattainable levels of discrimination, resulting in the accumulation of direct evidence of germ-cell mutagenesis in man. In order to meet the regulatory challenges this will present, Dr. DeMarini advocates for an IARC-type, international approach to the assessment of potential human germ-cell mutagens. Environ. Mol.
